The mRNA-encoded constitutively active STING variant is a novel mRNA-based therapeutic designed to overcome limitations of conventional STING agonists in cancer treatment. It encodes a constitutively active variant of Stimulator of interferon genes (STING) with an optimized CDN-binding domain to enhance signaling potency. Upon administration, this engineered mRNA leads to robust activation of the STING/TBK1/IRF3 pathway, triggering the production of type I interferons and proinflammatory cytokines, which are crucial mediators of anti-tumor immunity. Preclinical studies in syngeneic mouse models (MC38 and LL/2) have demonstrated its ability to significantly suppress tumor growth, highlighting its potent immunotherapeutic potential.
