mRNA encoding human cytomegalovirus UL128 is a specific messenger RNA component of Moderna's cytomegalovirus (CMV) vaccine candidate, **mRNA-1647**. It encodes the UL128 protein, which is one of the five subunits (along with gH, gL, UL130, and UL131A) that form the CMV pentameric complex. This complex is essential for the virus's ability to infect epithelial, endothelial, and myeloid cells. When administered as part of the lipid nanoparticle-encapsulated mRNA-1647 vaccine, this mRNA is translated by the host's cells to produce the UL128 antigen. This antigen, in conjunction with the other pentamer subunits and glycoprotein B (gB), elicits a robust neutralizing antibody response. The primary goal of this therapy is to prevent primary CMV infection in seronegative individuals, particularly to reduce the risk of congenital CMV transmission during pregnancy.
