MRT-5410 is an mRNA-based vaccine candidate originally developed for the prevention of COVID-19. It was the lead candidate from a collaboration between Sanofi and Translate Bio, utilizing Translate Bio's proprietary mRNA platform and lipid nanoparticle (LNP) delivery technology. The vaccine works by delivering messenger RNA that encodes the prefusion-stabilized spike protein of the SARS-CoV-2 virus. Once injected intramuscularly, host cells take up the LNPs and translate the mRNA into the spike protein, which then triggers an adaptive immune response, including the production of neutralizing antibodies and T-cell activation. Although the candidate demonstrated positive interim results in Phase 1/2 clinical trials, Sanofi announced in September 2021 that it would discontinue the development of MRT-5410 to focus on its recombinant protein-based vaccine and next-generation mRNA efforts, citing the sufficient supply of existing mRNA vaccines in the market.
