Overview

MRT-55811 is a potent and selective small molecule inhibitor of ubiquitin-specific protease 7 (USP7), a deubiquitinating enzyme that plays a critical role in regulating protein stability within the cell. Developed by Mission Therapeutics, MRT-55811 aims to treat various cancers by modulating the p53 pathway. USP7 typically stabilizes MDM2, an E3 ligase that targets the tumor suppressor p53 for degradation. By inhibiting USP7, MRT-55811 promotes the auto-ubiquitination and degradation of MDM2, thereby stabilizing p53 and inducing p53-mediated cell cycle arrest and apoptosis in tumor cells. Beyond the p53 axis, USP7 inhibition may also impact other oncogenic drivers and the tumor immune microenvironment. MRT-55811 has been utilized in preclinical research to demonstrate the therapeutic potential of DUB inhibition in oncology.
