MRT-8102 is a potent, highly selective, and orally bioavailable investigational small molecule classified as a molecular glue degrader (MGD) that targets NEK7 (NIMA-related kinase 7). By promoting the degradation of NEK7, MRT-8102 inhibits the assembly and activation of the NLRP3 inflammasome complex and reduces production of pro-inflammatory cytokines such as interleukin-1β (IL-1β) and interleukin-18 (IL-18). Aberrant activation of this pathway is implicated in multiple inflammatory diseases including gout, cardiovascular disease, neurodegenerative disorders like Parkinson’s disease and Alzheimer’s disease, ocular diseases, diabetes, obesity, and liver disease. Preclinical studies in non-human primates have shown that MRT-8102 can cross the blood-brain barrier to enter the brain and achieve near-complete reductions in IL-1β with a favorable safety profile. The drug is being developed by Monte Rosa Therapeutics for inflammatory conditions driven by IL-1β/NLRP3 inflammasome activity[1][5][6][7].
