MRT5400 is an investigational mRNA-based influenza vaccine formulation developed by Sanofi and Translate Bio that encodes the hemagglutinin (HA) antigen from a seasonal A/H3N2 influenza virus strain and is delivered in a proprietary lipid nanoparticle (LNP) composition. It represents one of two closely related monovalent mRNA vaccine formulations (MRT5400 and MRT5401) that share the same HA-encoding mRNA but differ in the LNP used to encapsulate and deliver the mRNA to host cells, where it is translated into HA to elicit neutralizing antibody and T-cell responses against influenza A/H3N2.[1][4][7][9] Preclinical studies in non-human primates showed that the MRT5400 LNP formulation induced robust functional antibody responses and T-cell cytokine responses, supporting advancement into a Phase 1 clinical trial in healthy adults to evaluate safety, tolerability, and immunogenicity as a next-generation seasonal influenza vaccine approach.[5][7][8]
