Overview

MRT5421 is a potent and selective molecular glue degrader (MGD) developed by Monte Rosa Therapeutics that targets the translation termination factor G1 to S phase transition 1 (GSPT1). As a molecular glue, MRT5421 facilitates the recruitment of GSPT1 to the Cereblon (CRBN) E3 ubiquitin ligase complex, inducing the polyubiquitination and subsequent proteasomal degradation of the target protein. GSPT1 is essential for protein synthesis and is frequently overexpressed in various malignancies, particularly those driven by the MYC oncogene. The degradation of GSPT1 by MRT5421 leads to the inhibition of protein translation, induction of the integrated stress response, and promotion of apoptosis in cancer cells. MRT5421 is part of a broader pipeline of GSPT1 degraders, including the clinical-stage candidate MRT-2359, and is being explored for its potential in treating advanced solid tumors.
