Overview

MS-C19 is a small molecule inhibitor of fatty acid synthase (FASN), derived from the natural product platensimycin. It is being investigated for the treatment of myeloid leukemia, specifically acute myeloid leukemia (AML). By targeting FASN, MS-C19 suppresses leukemic cell growth, survival, and clonogenicity while inducing myeloid differentiation. Mechanistically, inhibition of FASN by MS-C19 triggers the activation of lysosomal and inflammatory signaling pathways, including the upregulation of Progranulin. In preclinical models, MS-C19 has demonstrated superior potency compared to other FASN inhibitors like Orlistat and TVB-3166.
