MS-IL-15 is a long-acting interleukin-15 (IL-15) prodrug developed by ProLynx Inc. for cancer immunotherapy. The therapeutic consists of the cytokine IL-15 covalently attached to tetra-polyethylene glycol (tetra-PEG) hydrogel microspheres (MS) via a self-cleaving beta-eliminative linker. This formulation creates a localized drug depot upon subcutaneous or intratumoral administration, allowing for the slow, controlled release of active IL-15 with an initial half-life of approximately 7 days. MS-IL-15 functions as an agonist of the IL-2 receptor beta/gamma (IL-2Rβ/γc) complex, which is expressed on natural killer (NK) cells, CD8+ T cells, and gamma-delta T cells. By providing sustained exposure to IL-15, the drug promotes the prolonged expansion and activation of these cytotoxic immune populations without significantly inducing immunosuppressive regulatory T cells (Tregs). Preclinical studies have demonstrated its efficacy in various tumor models, including prostate cancer, adult T-cell leukemia, and triple-negative breast cancer, particularly when used in combination with other immunotherapeutic agents.
