MS-RLI is a long-acting research prodrug consisting of the IL-15 agonist RLI (receptor-linker-IL-15) conjugated to tetra-PEG hydrogel microspheres (MS). RLI is a fusion protein comprising human IL-15 covalently linked to the human IL-15Rα sushi+ domain, designed to act as a potent IL-15 superagonist. The MS-RLI conjugate utilizes a releasable linker that allows for the sustained release of the active RLI moiety via a pre-programmed β-elimination reaction, effectively extending the drug's half-life and creating a localized depot. This technology, developed by ProLynx, is intended to stimulate the expansion and activation of natural killer (NK) cells and CD8+ T cells while minimizing the systemic toxicities often associated with IL-15 agonists. Preclinical studies have evaluated MS-RLI for the treatment of solid tumors, such as colon carcinoma and triple-negative breast cancer, demonstrating significant anti-metastatic activity when administered intratumorally.
