Overview

The mSA2 affinity-enhanced biotin-binding CAR is a universal anti-tag chimeric antigen receptor (AT-CAR) cell therapy. It utilizes an engineered monomeric streptavidin 2 (mSA2) domain that has been optimized for high-affinity binding to biotin. Unlike traditional CAR-T cells that target a specific tumor antigen directly, mSA2 CAR-T cells are designed to bind to biotinylated tumor-specific antibodies. This modular approach allows the same CAR-T cell population to be redirected against various tumor-associated antigens (such as CD19 or CD20) simply by changing the biotinylated antibody administered. The construct typically incorporates signaling domains such as CD28-CD3ζ or 4-1BB-CD3ζ to trigger T-cell activation and tumor lysis upon binding to the biotin-tagged antibodies on the cancer cell surface. This technology aims to provide a flexible platform to overcome antigen escape and enable multi-antigen targeting.
