Drug intelligence / Profile preview
mSA2 CAR-T cells are an adaptable, universal anti-tag chimeric antigen receptor (CAR) T-cell therapy platform designed to overcome tumor heterogeneity and antigen escape. Instead of a traditional single-chain variable fragment (scFv) targeting a specific tumor antigen, these CAR-T cells express an affinity-enhanced monomeric streptavidin-2 (mSA2) biotin-binding domain. The mSA2 domain binds with high affinity to biotinylated tumor-targeting antibodies (such as those targeting CD19, CD20, CD276, EPHA2, CD70, or IL13Rα2), which are co-administered to direct the T-cells to the tumor site. This modular approach allows a single population of CAR-T cells to target multiple antigens simultaneously or sequentially, and provides a safety mechanism where CAR-T cell activity can be controlled by adjusting the dose of the biotinylated antibody. The platform was originally developed at the University of Pittsburgh and is licensed to Monarch Therapeutics, with academic research also being conducted by the German Cancer Research Center (DKFZ) and Heidelberg University Hospital for indications like glioblastoma.
