MSAB (Methyl 3-(4-methylbenzenesulfonamido)benzoate) is a small molecule inhibitor that selectively targets the Wnt/β-catenin signaling pathway. It functions by directly binding to the C-terminal armadillo repeats of β-catenin, which promotes its degradation through the ubiquitin-proteasome system. By reducing the levels of active β-catenin, MSAB inhibits the transcription of Wnt target genes such as CCND1 (cyclin D1) and MYC, which are essential for tumor cell proliferation and survival. Preclinical research has demonstrated that MSAB can effectively suppress tumor growth in various models, including colorectal and prostate cancer. Furthermore, it has been shown to attenuate cancer stemness and overcome resistance to chemotherapy agents like docetaxel in advanced prostate cancer cells.
