MSC-L is an engineered cell therapy product consisting of human umbilical cord-derived allogenic mesenchymal stem cells (MSCs) that have been genetically modified to express an antitumor CXCL-family chemokine and a TNF-superfamily co-stimulatory molecule. Developed by Shanghai East Hospital for the treatment of advanced colorectal cancer, MSC-L is designed to leverage the natural tumor-homing capabilities of MSCs to deliver therapeutic proteins directly into the tumor microenvironment. The expressed chemokines are intended to recruit peripheral T cells and natural killer (NK) cells to the tumor, while the co-stimulatory molecules provide the necessary signals to activate these infiltrated lymphocytes, thereby promoting a robust and persistent antitumor immune response. This strategy is particularly aimed at treating "cold" tumors with unfavorable immune microenvironments that typically respond poorly to standard immune checkpoint inhibitors.
