MSC-NTC (Mesenchymal Stem Cell - Non-Targeting Control) is an engineered cell population used as a negative control in preclinical studies evaluating mesenchymal stem cell (MSC)-based therapies. These cells are typically modified with a non-targeting genetic sequence (such as a scrambled shRNA or a non-functional microRNA) to serve as a baseline for comparing the effects of MSCs engineered with specific therapeutic payloads, such as miR-379. MSCs are utilized in these contexts due to their innate ability to home to tumor sites. In studies of metastatic breast cancer, MSC-NTC is used to demonstrate that observed therapeutic effects are due to the specific genetic modification rather than the MSC vehicle itself. Preclinical data indicates that while MSC-NTC cells home to tumors, they do not significantly impact tumor growth or formation compared to therapeutic variants.
