Overview

MSC-oHSV is an experimental cell-based oncolytic therapy consisting of mesenchymal stem cells (MSCs) loaded with an oncolytic herpes simplex virus (oHSV). This approach utilizes the innate tumor-homing capabilities of MSCs to deliver the viral payload directly to metastatic sites, including those in the brain, thereby overcoming barriers like virus neutralization and poor extravasation associated with systemic viral delivery. Once at the tumor site, the oHSV selectively replicates within and lyses tumor cells, releasing neoantigens and stimulating an anti-tumor immune response. Research conducted at Brigham and Women's Hospital and Massachusetts General Hospital has demonstrated its efficacy in preclinical models of advanced melanoma and melanoma brain metastases, particularly when combined with immunomodulators like GM-CSF.
