Overview

MSC supernatant refers to the cell-free culture medium collected after mesenchymal stem cells (MSCs) have been cultured. This supernatant contains a complex mixture of bioactive molecules, including growth factors, cytokines, chemokines, and extracellular vesicles such as exosomes. These components are secreted by MSCs and mediate many of their therapeutic effects through paracrine signaling rather than direct cell replacement. The MSC supernatant has demonstrated regenerative, anti-inflammatory, immunomodulatory, pro-angiogenic, and wound-healing properties in preclinical studies. It is being explored as an alternative to live-cell therapy due to its ease of production, storage (including lyophilized forms), reduced safety concerns (e.g., no risk of tumor formation or embolism), and potential for large-scale manufacturing[2][3][6]. The mechanism of action is primarily via modulation of immune responses and promotion of tissue repair through the delivery of soluble factors that influence target cells' behavior[3][6]. Sources for MSCs include bone marrow, adipose tissue, umbilical cord blood/placenta among others[2][6].
