MSK-777 is a novel natural product-derived small molecule inhibitor of Cdc7 kinase, developed at Memorial Sloan-Kettering Cancer Center. Cdc7 is a serine-threonine kinase essential for the initiation of DNA replication and the S-phase checkpoint. Inhibition of Cdc7 by MSK-777 leads to cell cycle arrest at the G1/S transition, accumulation of DNA damage, and induction of apoptosis in various cancer types, including pancreatic carcinoma, solid tumors, and hematologic malignancies. Preclinical studies have demonstrated its efficacy in reducing cell viability and inhibiting the phosphorylation of Mcm2, a downstream target of Cdc7, in human pancreatic cancer cell lines and animal models.
