Overview

The MSKCC multivalent conjugate vaccine is a hexavalent cancer immunotherapy biologic developed by Memorial Sloan Kettering Cancer Center for the treatment of biochemically relapsed prostate cancer. The vaccine is composed of six synthetic tumor-associated antigens—GM2, Globo H, Lewis y, glycosylated MUC-1 (32mer), Tn, and TF—each individually conjugated to the carrier protein Keyhole Limpet Hemocyanin (KLH). This formulation is administered subcutaneously alongside the saponin-based adjuvant QS-21 (or alternatively GPI-0100). The vaccine is designed to overcome immunologic tolerance and induce a robust antibody response against carbohydrate and glycoprotein targets that are overexpressed on the surface of prostate cancer cells. In Phase II pilot studies, the vaccine demonstrated safety and the ability to break tolerance against multiple antigens in all vaccinated patients, although antibody titers were generally lower than those observed with monovalent versions of the individual components.
