Drug intelligence / Profile preview
MSL-7 is a small-molecule autophagy enhancer and a chemically modified derivative of MSL with improved microsomal stability. It acts by activating calcineurin, which dephosphorylates and promotes the nuclear translocation of the transcription factors TFEB and TFE3, master regulators of lysosomal biogenesis and autophagy gene expression. This activation occurs in an mTORC1-independent manner. MSL-7 has been shown to enhance lipophagy, reduce ectopic lipid accumulation, and decrease metabolic inflammation in obesity and diabetes models. Additionally, it promotes the clearance of toxic human islet amyloid polypeptide (hIAPP) oligomers and fibrils, protecting pancreatic beta-cells from apoptosis and improving glucose tolerance and beta-cell function in vivo.
