Overview

MSLN ADC refers to any antibody-drug conjugate (ADC) specifically designed to target mesothelin (MSLN), a glycosylphosphatidylinositol-anchored cell surface protein highly expressed in several solid tumors such as ovarian carcinoma, lung adenocarcinoma, and mesothelioma. These ADCs consist of a monoclonal antibody that binds selectively to mesothelin on tumor cells, linked via a chemical linker to a cytotoxic payload. Upon binding to the tumor cell surface antigen (mesothelin), the complex is internalized and trafficked into lysosomes where the linker is cleaved, releasing the cytotoxic agent inside the cancer cell and inducing apoptosis or other forms of cell death. The mechanism leverages both targeted delivery and potent cytotoxicity for improved efficacy with reduced off-target toxicity compared to conventional chemotherapy[1][2][3][5]. Multiple MSLN-targeted ADCs have been developed or are in development by various companies using different antibodies and payloads; notable examples include anetumab ravtansine (Bayer) and HBM9033 (Nona Biosciences)[3].
