Overview

MSLN CAR-T TGFβRIIDN is an experimental mesothelin-targeted chimeric antigen receptor T cell therapy that has been further engineered with a dominant-negative transforming growth factor beta receptor II (TGFβRII-DN) to resist TGF-β–mediated immunosuppression within the tumor microenvironment. Mesothelin is a tumor-associated glycoprotein highly expressed on various solid tumors, including mesothelioma, pancreatic, ovarian, and certain lung cancers, and serves as an attractive target for CAR-T therapy, though on-target, off-tumor risks exist because of its low-level expression on normal mesothelial tissues.[3][8] By combining MSLN-directed CAR-T cells with a TGFβRIIDN “armoring” strategy that blocks inhibitory TGF-β signaling in the infused T cells, this product is designed to enhance proliferation, persistence, and cytotoxic function of CAR-T cells in solid tumors, a concept supported by emerging dnTGFβRII-armored CAR-T platforms that have shown improved responses in solid tumor settings.[4]
