MSLN-Pep-KLA is a chimeric proapoptotic peptide designed for the targeted treatment of mesothelin-overexpressing malignancies, particularly pancreatic cancer. It consists of a 9-mer mesothelin-binding peptide (MSLN-Pep), identified via phage display, conjugated to a proapoptotic KLA peptide (typically (KLAKLAK)2). The MSLN-Pep moiety facilitates selective binding and internalization into cancer cells expressing high levels of mesothelin, a cell-surface glycoprotein frequently overexpressed in pancreatic and ovarian cancers. Once internalized, the KLA peptide disrupts mitochondrial membranes, inducing selective cytotoxicity and apoptosis. Preclinical research indicates that MSLN-Pep-KLA can inhibit cell migration and invasion and may act synergistically with chemotherapeutic agents like gemcitabine to enhance anti-tumor efficacy.
