MSLN STAR-T cells are autologous T cells genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets mesothelin (MSLN), a glycoprotein overexpressed in many solid tumors such as ovarian, lung, and mesothelioma. The "STAR" in the name refers to a specific design or tuning of the CAR construct to optimize activation potential and persistence. These modified T cells are infused intravenously into patients with advanced MSLN-positive solid tumors. The therapy aims to enhance antitumor activity while reducing T cell exhaustion by calibrating CAR signaling domains—such as using a mutated CD3ζ chain containing a single ITAM—to improve efficacy and durability in the immunosuppressive tumor microenvironment[1][2][3][5]. This approach is under investigation for its safety and effectiveness in clinical trials.
