MSLNxEpCAM Switch-DARPin is a preclinical multi-specific DARPin-based T‑cell engager engineered by Molecular Partners to target solid tumors, particularly ovarian cancer, that co-express mesothelin (MSLN) and epithelial cell adhesion molecule (EpCAM). It incorporates DARPin domains specific for MSLN and EpCAM together with a CD3-binding DARPin that is “masked” via a switch design, allowing conditional CD3 engagement and T‑cell activation only in the presence of both tumor-associated antigens, thereby aiming to enhance tumor specificity and reduce off-tumor toxicity and cytokine release.[1][2] The molecule is part of Molecular Partners’ Switch-DARPin platform, which provides logic-gated on/off control of immune cell engagers to improve safety and potency in solid tumors.[1][2]
