MST-997 is an antibody-drug conjugate (ADC) that was developed by Wyeth (now a wholly owned subsidiary of Pfizer) for the treatment of advanced malignant solid tumors, including metastatic breast cancer and non-small cell lung cancer. The agent consists of a humanized monoclonal antibody (huHMFG1) targeting Mucin 1 (MUC1), a cell surface glycoprotein that is frequently overexpressed and aberrantly glycosylated in various epithelial malignancies. The antibody is conjugated to the cytotoxic maytansinoid derivative DM1 (mertansine), a potent microtubule inhibitor. Upon binding to MUC1 on the surface of cancer cells and subsequent internalization, the DM1 payload is released, leading to cell cycle arrest in the G2/M phase and apoptosis. Clinical development reached Phase 1 clinical trials (NCT00094133) but was subsequently discontinued.
