MSV + proDox (also referred to as MSV/proDox or proDox in MSV) is an experimental nanotherapeutic formulation consisting of a doxorubicin prodrug (proDox) loaded into a silicon-based multistage vector (MSV) system [5697]. Developed by researchers at the Houston Methodist Research Institute, this platform is designed to overcome biological barriers and selectively deliver chemotherapeutic agents to tumor tissues, such as breast cancer lung metastases, while minimizing systemic and cardiac toxicity [5697]. The MSV carrier consists of mesoporous silicon microparticles that protect the prodrug cargo during circulation. Upon reaching the tumor microenvironment, the doxorubicin prodrug is cleaved and activated to release active doxorubicin, which intercalates into DNA and inhibits topoisomerase II, leading to cancer cell death.
