MT-125 is a first-in-class, selective small molecule inhibitor of non-muscle myosin II (NMII), a molecular motor protein implicated in tumor proliferation, invasion, resistance to therapy, oxidative stress response, and immune evasion. Developed by Myosin Therapeutics in collaboration with the Mayo Clinic and Scripps Research/University of Florida, MT-125 is designed to disrupt critical mechanisms driving glioblastoma progression. Preclinical studies have shown that MT-125 has potent anti-tumor activity and enhances the efficacy of radiotherapy. The drug has received FDA Orphan Drug Designation for malignant gliomas (including glioblastoma) and is entering Phase 1 clinical trials for newly diagnosed IDH wild type, MGMT unmethylated glioblastoma[6][3][5][2].
