Overview

MT-302 is a first-in-class, mRNA-based in vivo chimeric antigen receptor (CAR) therapy designed to target TROP2-expressing epithelial tumors. Unlike traditional CAR-T therapies that require ex vivo cell manipulation, MT-302 uses synthetic mRNA encapsulated in lipid nanoparticles (LNPs) to program myeloid cells directly within the patient’s body. The therapy induces these myeloid cells to express a CAR specific for TROP2, a tumor-associated antigen overexpressed in many solid epithelial cancers but with limited expression in normal tissues. This approach enables direct cytotoxicity against tumor cells and triggers an adaptive immune response by presenting tumor neoantigens, potentially leading to long-term immune surveillance and reduced risk of recurrence. MT-302 is being developed by Myeloid Therapeutics and is currently undergoing Phase 1 clinical trials for multiple advanced or metastatic epithelial tumors[2][3][5][7].
