MT-4019

**MT-4019** is a preclinical engineered toxin body (ETB) developed by Molecular Templates targeting **CD38**, a protein highly expressed on multiple myeloma cells. It represents the company's second-generation ETB platform featuring a proprietarily de-immunized scaffold to reduce immunogenicity while delivering a modified bacterial toxin that enzymatically and irreversibly destroys ribosomes inside target cells, leading to cell death independent of the immune system. Initially planned for Phase 1 initiation in relapsed/refractory multiple myeloma in 3Q2018 with $15.3M CPRIT grant funding, the program was advanced through a collaboration with Takeda, yielding more potent CD38-targeted ETBs intended for clinical development in oncology, particularly multiple myeloma.
