MT-7716 is a **novel, selective, nonpeptidergic agonist of the nociceptin/orphanin FQ (NOP) receptor**. It is a small molecule designed to **cross the blood-brain barrier** and acts as a full agonist at the NOP receptor with high affinity. MT-7716 does **not activate mu, delta, or kappa opioid receptors**, distinguishing it from classical opioids[4][2][1][5]. The main pharmacological effect is to **decrease GABAergic transmission in the central amygdala (CeA)**, effectively blocking ethanol-induced increases in GABA release. Preclinical studies demonstrated that MT-7716 **reduces alcohol self-administration, prevents stress and alcohol cue-induced relapse, and attenuates alcohol withdrawal symptoms in rats**[3][2][4][5][6]. Its therapeutic potential was explored for the treatment of **alcohol use disorder (alcoholism)**[8]. Development was discontinued after Phase I clinical trials[8].
