MT1-MMP bicycle drug conjugates (BDCs) are a class of peptide-drug conjugates developed by Bicycle Therapeutics. They consist of a constrained bicyclic peptide (Bicycle®) that binds with high affinity to the hemopexin domain of Membrane Type 1-Matrix Metalloproteinase (MT1-MMP, also known as MMP14), linked to a cytotoxic payload via a cleavable linker. The platform includes specific candidates such as BT1718, which carries the maytansinoid tubulin inhibitor DM1, and BT1769, which carries monomethyl auristatin E (MMAE). These conjugates are designed to penetrate tumors rapidly due to their low molecular weight (1.5-3 kDa) and exhibit a short systemic half-life, potentially reducing off-target toxicities compared to traditional antibody-drug conjugates (ADCs). MT1-MMP is highly expressed in various solid tumors, including non-small cell lung cancer (NSCLC), triple-negative breast cancer, and sarcomas, making it a viable target for precision delivery of toxins.
