Overview

MT114 is a bispecific T-cell engager (BiTE) antibody designed for the treatment of acute myeloid leukemia (AML). It simultaneously binds to the CD33 (SIGLEC-3) antigen, which is frequently expressed on AML blasts and leukemic stem cells, and the CD3 epsilon subunit of the T-cell receptor complex. By bridging these two targets, MT114 facilitates the redirected lysis of CD33-positive tumor cells by unstimulated T cells, leading to T-cell activation and the release of inflammatory cytokines such as IFN-gamma, TNF-alpha, and various interleukins. Developed by Micromet (later acquired by Amgen), MT114 was engineered to recognize epitopes conserved between humans and macaques to facilitate preclinical safety and pharmacological testing. It demonstrated potent redirected lysis of multiple human AML cell lines in preclinical studies and showed efficacy in delaying tumor growth in HL60 xenograft models.
