MT1988

**MT1988** is an investigational, orally administered fixed-dose combination of the small molecules tropisetron and varenicline, developed by Monument Therapeutics for cognitive impairment associated with schizophrenia. The combination is intended to enhance nicotinic cholinergic signalling without dopamine D2 receptor blockade: tropisetron provides partial agonism at alpha-7 nicotinic acetylcholine receptors and antagonism of 5-hydroxytryptamine receptor 3, while varenicline provides agonism at alpha-7 and alpha-4/beta-2 nicotinic acetylcholine receptors and agonism at 5-hydroxytryptamine receptor 3. MT1988 completed a Phase 1 healthy-volunteer study and is being evaluated in an eight-week proof-of-principle study in people at clinical high risk for psychosis, using cognitive, clinical, biological, and digital biomarkers.
