MT218 is a small peptide-based targeted MRI contrast agent designed for the accurate early detection and risk stratification of aggressive solid tumors. It consists of a seven-amino-acid peptide (ZD2) conjugated to the clinical macrocyclic gadolinium-based contrast agent Gd(HP-DO3A). The peptide specifically targets extradomain B fibronectin (EDB-FN), an oncofetal variant of fibronectin that is highly expressed in the extracellular matrix of aggressive cancers but not in normal tissues. By binding to EDB-FN, MT218 enables robust and specific magnetic resonance imaging (MRI) enhancement at lower doses than conventional gadolinium agents. This allows for sensitive detection and characterization of tumor invasiveness, assessment of drug resistance, and monitoring response to therapy across multiple cancer types including breast cancer, colorectal cancer, head and neck cancer, lung cancer, pancreatic cancer, and high-risk prostate cancer.
