Overview

MT3014 is a selective small-molecule inhibitor of phosphodiesterase 10A (PDE10A), an enzyme that hydrolyzes the cyclic nucleotides cAMP and cGMP. Developed by Mitsubishi Tanabe Pharma, MT3014 has been primarily advanced through Phase 2 clinical trials for the treatment of schizophrenia, where it aims to modulate dopaminergic and glutamatergic signaling in the striatum to alleviate both positive and negative symptoms. Recent preclinical research, such as that presented at the AHA 2025 meeting, has also demonstrated its potential in treating right ventricular (RV) dysfunction. In models of pulmonary artery banding, MT3014-mediated inhibition of PDE10A was found to attenuate RV remodeling and hypertrophy by modulating TRPV2-mediated calcium signaling and the β-catenin pathway, suggesting a novel therapeutic application for heart failure.
