Drug intelligence / Profile preview
MTA-304 (formerly EM-221) is a highly selective, second-generation small molecule phosphodiesterase 10A (PDE10A) inhibitor being developed by Mentara Therapeutics. Originally discovered by Mochida Pharmaceutical and previously developed by EuMentis Therapeutics, the compound is designed to target levodopa-induced dyskinesia (LID) in patients with Parkinson's disease. By inhibiting PDE10A, which is highly expressed in the striatum, MTA-304 aims to rebalance striatal signaling pathways overstimulated by levodopa, thereby reducing involuntary movements without diminishing levodopa's therapeutic benefit. MTA-304 has completed three Phase 1 clinical trials demonstrating a favorable safety and tolerability profile, and is planned to enter Phase 2 clinical development in the first half of 2027.
