mTa-CAR-T cells are engineered T cells designed to target the tumor microenvironment (TME) and induce Tertiary Lymphoid Structures (TLS). These cells are constructed with a membrane-tethered anti-Lymphotoxin-beta receptor (LTβR) single-chain variable fragment (Sc-Fv) alongside a Chimeric Antigen Receptor (CAR) construct. By functioning as lymphoid tissue inducer cells (LTi) and interacting with lymphoid tissue organizer cells (LTo) that express LTβR, mTa-CAR-T cells aim to nucleate TLS within aggressive tumors like glioblastoma multiforme (GBM). The induction of TLS is hypothesized to make the TME more immune-permissive, thereby enhancing the efficacy of CAR-T therapy and improving patient survival.
