Overview

MTA2-K539la targeting peptide is an engineered cell-penetrating peptide designed to disrupt the lactylation of Metastasis-associated protein 2 (MTA2) at the lysine 539 (K539) residue. MTA2 is a core component of the Nucleosome Remodeling and Deacetylase (NuRD) complex, which plays a critical role in DNA damage repair. In lung adenocarcinoma (LUAD), the Warburg effect leads to lactate accumulation, which triggers KAT2A-mediated lactylation of MTA2 at K539. This post-translational modification enhances the binding affinity of MTA2 to importin α, promoting its nuclear translocation and subsequent stabilization of the NuRD complex. This mechanism suppresses cisplatin-induced DNA damage, contributing to chemoresistance. By specifically targeting this lactylation site, the peptide aims to inhibit the nuclear translocation of the NuRD complex, thereby resensitizing refractory LUAD tumors to conventional chemotherapeutic and immunotherapeutic agents.
