MTADV 5-mer peptide (5MP) is a synthetic pentapeptide with the sequence Methionine-Threonine-Alanine-Aspartic Acid-Valine, derived from a specific segment of the CD44 variant protein. It is being investigated as an anti-inflammatory therapeutic that targets and inhibits the pathological forms of Serum Amyloid A (SAA). The peptide works by binding to SAA and interfering with its polymerization and aggregation, processes that drive chronic inflammation and amyloidosis. By inhibiting SAA activity, 5MP suppresses the release of pro-inflammatory cytokines, including IL-6, IL-1β, and TNFα, from innate immune cells. Developed by researchers at the Hebrew University of Jerusalem, the peptide has shown efficacy in preclinical models of inflammatory bowel disease (IBD), multiple sclerosis (MS), and rheumatoid arthritis (RA), and has demonstrated a favorable safety profile in canine studies.
