Overview

**Mtb72F** is a recombinant fusion protein subunit vaccine candidate developed for the prevention of tuberculosis (TB). It is composed of regions from two *Mycobacterium tuberculosis* proteins: Mtb32 (a serine protease, Rv0125) and Mtb39 (a PPE family protein, Rv1196). Mtb72F is designed to stimulate strong cellular immune responses, primarily inducing CD4+ T cell proliferation and interferon-gamma (IFN-γ) production, leading to enhanced protection against TB. The vaccine has been shown to provide greater immunogenicity and protection in animal models compared to its individual components. Mtb72F has also been formulated as a DNA vaccine and in combination with adjuvants such as AS02 in clinical trials. Its main clinical development as a protein subunit vaccine has focused on use as a booster after BCG or as a primary vaccine, with particular relevance in immunocompromised patients. It is being tested both as a recombinant protein vaccine (Mtb72F/AS02) and as a DNA vaccine (pcDNA3.1-Mtb72F).
