MTD217 is a preclinical-stage combination therapy developed by Biodexa Pharmaceuticals (formerly Midatech Pharma) for the treatment of leptomeningeal disease (LMD), a secondary metastatic cancer of the central nervous system. The drug candidate is a combination of MTX110, which is a solubilized liquid formulation of the histone deacetylase (HDAC) inhibitor panobinostat, and an oxidative phosphorylation (OXPHOS) inhibitor. MTD217 is designed to simultaneously target two primary metabolic pathways used by cancer cells for energy production: aerobic glycolysis (the Warburg effect) and mitochondrial oxidative phosphorylation. By inhibiting both pathways, the therapy aims to overcome metabolic plasticity and prevent cancer cells from adapting to cellular stress. The formulation utilizes the proprietary MidaSolve nanotechnology platform to solubilize otherwise insoluble drugs, enabling direct delivery into the cerebrospinal fluid or tumor microenvironment to maximize local therapeutic concentration while minimizing systemic off-target toxicity.
