MTH1 siRNA is a small interfering RNA designed to silence the expression of MutT homolog 1 (MTH1), also known as NUDT1. MTH1 is an enzyme that hydrolyzes oxidized purine nucleoside triphosphates (such as 8-oxo-dGTP), preventing their incorporation into DNA and subsequent DNA damage-induced apoptosis. In cancer cells, which often exhibit high levels of reactive oxygen species (ROS), MTH1 is frequently upregulated to maintain genomic integrity. Research, particularly from the University Hospital Cologne, has demonstrated that siRNA-mediated knockdown of MTH1 in glioblastoma multiforme (GBM) cells leads to impaired cell migration and reduced tumor growth in vivo.
