Overview

MTI-301 is an orally bioavailable small-molecule inhibitor of stearoyl-CoA desaturase 1 (SCD1), a key enzyme involved in fatty acid synthesis. SCD1 activity is upregulated in several aggressive cancers and contributes to tumor growth, metastasis, and drug resistance by promoting the synthesis of monounsaturated fatty acids. By inhibiting SCD1, MTI-301 aims to disrupt these processes and reduce tumor progression. The drug is being developed by Modulation Therapeutics for the treatment of advanced malignancies, including metastatic or unresectable solid tumors that are refractory to standard therapies. A first-in-human Phase 1 clinical trial sponsored by Mayo Clinic is underway to evaluate its safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced cancer[1][2][3][4][7]. Preclinical data also suggest potential for combination with immune checkpoint inhibitors[10].
