Overview

MTK458 is a small molecule activator of **PTEN induced kinase 1 (PINK1)**, a mitochondrial kinase that plays a central role in mitochondrial quality control. PINK1 identifies damaged mitochondria and initiates **mitophagy**, the selective degradation of dysfunctional organelles. Mutations in the PINK1 gene are a known cause of autosomal recessive early-onset Parkinson's disease, and PINK1 deficiency is implicated in the pathogenesis of idiopathic Parkinson's. MTK458 is designed to potently and selectively enhance PINK1 activity, thereby promoting the clearance of damaged mitochondria and protecting neurons from oxidative stress and proteotoxicity. Originally developed by **Mitokinin**, the program was acquired by **AbbVie** in 2023 to advance it toward clinical development. Preclinical studies have demonstrated that MTK458 can rescue locomotion defects and neurite length in various Parkinson's disease models, including those characterized by UQCRC1 deficiency.
