Overview

MTL-004 is a small molecule therapeutic being developed by Gordian Pharma and Oncodesign Services for the treatment of glioblastoma (GBM). Chemically identified as 5-(aziridine-1-yl)-4-hydroxylamino-2-nitrobenzamide, it is a DNA alkylating agent designed for local delivery into the brain cavity following surgical resection of primary tumors. The drug is formulated in a bioresorbable paste composed of poly DL-lactic acid-co-glycolic acid (PLGA) and poly ethylene glycol (PEG), which acts as a sustained-release implant. This localized intracranial administration aims to control tumor recurrence and improve survival while minimizing the systemic toxicities associated with standard oral therapies. Preclinical studies in rat glioma models have demonstrated that a single intracranial dose of MTL-004 significantly increases survival compared to the standard-of-care oral temozolomide.
