This is a combination therapy consisting of an mTOR inhibitor (such as sirolimus or a dual PI3K/mTOR inhibitor like NVP-BEZ235) and nanoparticle albumin-bound paclitaxel (nab-paclitaxel). The mTOR inhibitors act by blocking the mammalian target of rapamycin (mTOR), a key regulator of cell growth, proliferation, and survival. Nab-paclitaxel is a taxane chemotherapeutic agent formulated with albumin nanoparticles to enhance delivery and reduce toxicity compared to conventional paclitaxel. Preclinical studies have shown that combining an mTOR inhibitor with nab-paclitaxel can result in additive or synergistic antitumor effects, particularly through inhibition of cell proliferation, induction of apoptosis, and modulation of the PI3K/mTOR pathway. This combination has been investigated in solid tumors such as gastric cancer and breast cancer models[1][2][3][4].
