MTS108 is an mRNA-lipid nanoparticle (mRNA-LNP) therapeutic candidate designed for the treatment of small cell lung cancer (SCLC). It encodes a trispecific T-cell engager (TCE) that features a biparatopic design, utilizing two distinct camelid-derived VHH (nanobody) domains to target non-overlapping epitopes of Delta-like ligand 3 (DLL3). This biparatopic engagement is intended to increase avidity and improve tumor cell killing, particularly in tumors with low DLL3 expression. The TCE also includes a CD3 agonist domain to recruit and activate T cells against the tumor. Developed by Metis Pharmaceuticals, MTS108 utilizes an optimized mRNA-LNP platform to provide sustained in vivo expression of the TCE, aiming to improve the safety profile by reducing the risk of cytokine release syndrome (CRS) compared to traditional recombinant protein-based TCEs. Preclinical studies have demonstrated that MTS108 possesses potent anti-tumor activity, outperforming benchmarks such as xaluritamig in various lung cancer models.
