MTX-101 is a first-in-class, antigen-agnostic bispecific antibody developed as an autoimmune checkpoint inhibitor. It targets inhibitory KIR and CD8 expressed on regulatory CD8 T cells (CD8 Treg), aiming to restore their intrinsic functions early in the autoimmune disease process. By selectively activating regulatory CD8 T cells, MTX-101 suppresses and eliminates pathogenic CD4 T cells, halts downstream inflammation, and prevents tissue destruction without broad immunosuppression or unwanted immune cell activation. Preclinical studies have shown that MTX-101 increases the cytolytic capacity and prevalence of CD8 Tregs, reduces proinflammatory cytokines, and specifically targets autoreactive immune responses while sparing normal antiviral or antibacterial immunity. The drug is being developed for autoimmune diseases such as type 1 diabetes mellitus and celiac disease[1][2][3][4][5][6][8].
