Overview

MTX-474 is a first-in-class human IgG1 monoclonal antibody developed by Mediar Therapeutics. It is designed to neutralize EphrinB2 signaling, which plays a key role in the onset and progression of fibrosis. Ephrin ligands and their receptors are involved in biological processes such as cell migration, myofibroblast activation, and tissue remodeling—central mechanisms in fibrotic diseases. Preclinical evidence implicates EphrinB2 in the fibrosis of skin, lungs, and heart; its expression correlates with disease severity. MTX-474 is being developed primarily for systemic sclerosis (SSc) and other fibrosing conditions[1][2][3][4][7].
